What this category covers

Most compounds here act somewhere along the hypothalamic-pituitary-gonadal (HPG) axis, and their differences are best understood by where on that axis they act.

Upstream, at the hypothalamus: Kisspeptin-10 is studied as a regulator of GnRH release — a step above GnRH itself. At the GnRH receptor: Gonadorelin is synthetic GnRH with a short half-life, while Triptorelin is a longer-acting GnRH agonist. The half-life difference is not a minor detail: sustained GnRH-receptor agonism produces receptor downregulation, which is the opposite of the pulsatile stimulation short-acting analogs are studied for. Downstream, at the gonads: HCG acts on the LH receptor directly, and HMG supplies both FSH and LH activity.

PT-141 (Bremelanotide) sits outside the HPG axis entirely. It is a melanocortin-receptor agonist studied in central-nervous-system pathways, making it mechanistically closer to the melanocortin analogs Melanotan I and II than to the gonadotropins it is catalogued alongside.

The HPG axis responds to the pattern of stimulation, not just its presence.
